According to Virginia Tech's website, the word Hokie originated in 1896 from a spirit yell created by O.M. Stull, which is now known as the "Old Hokie." The school was changing its name from the Virginia Agricultural and Mechanical College to the Virginia Agricultural and Mechanical College and Polytechnic Institute and needed a new cheer that didn't reference the original, so Stull stepped in and won the competition. The first line of his chant contained the word "Hoki," but an "e" was added later.
